Really depends what place you're looking for, how much time you have to travel around Australia, how you want to get around, etc.?
If you'd ask a more specific question it would be easier to give you advice.

I don't think it matters where you start or end! As long as you get out of your trip what you want to. I personally didn't rate Cairns as a town much, although I stayed in the Serpent which was pretty nice - a little out of town but they provide regular shuttles etc. Did a wicked sky dive in cairns, and obviously the barrier reef etc. Never got to go to Cape Tribulation which i've been told is great. If you are arriving in the Aussie winter then its best to start up north and then travel back down to Sydney, Melbourne etc when its warmer down there.
Hope this has helped!!;)
