the two of us are traveling from Buenos Aires starting the end of this august, we have about 3 weeks to get to Bogota, Colombia and would like to see as much as possible without being rushed.
Any suggestion of routes to take, and definitely unmissable stops on this trip? we are looking less for wilderness adventures, and more to get to know places and cultures.
Would appreciate any suggestions we can get!

3 weeks from BA to Bogota you are going to be very rushed indeed, unless at some point in your trip you decide to take a flight.
I can't see how you will able to get much in 3 weeks traveling such a great distance.
Maybe you want to go via Iguazu Falls and then through to Florianapolis and Rio De J on your way. After that time a flight from Brazil to Bogota may be the best way to get there unless you want to go via Manaus and the Amazon, but 3 weeks is way too short a time for this type of journey.
I'm doing the same thing but starting in Venezuala before going onto Colombia and finishing in Rio after BA. Only difference, I'm going for 5 months.
Uno is right; this is not a feasible scenario, unless you want to be in a bus 24/7.
I would recommend that you revise your itinerary; consider flying back from Brasilia or Rio. The former still is pretty tight, but distance-wise at least, you can quite easily make it. Otherwise, taking a continental flight is the only solution, but take into account the enormous burden that puts on the environment.

thanks everyone, i am considering the flying option for part of the trip to gain some time. perhaps from Lima into Bogota, or from Buenos Aires into Lima. or out of Quito maybe.
but it seems that flights, even for short distances, are rather pricey in south america. any suggestions?
Out of Lima is very feasible; Quito's too far, will take you more time than you have to get there in a somewhat relaxed way.
There's a number of suggested itineraries in this forum for the BsAs-Lima stretch; consult those, or else I'd be happy to look up my own schedule from 3 years ago.
