Here's what you need to do:

El Bolson - Esquel - Comodoro Rivadavia - Rio Gallegos - Ushuaia

I did it some years back, and it was the most efficient option back then. You're likely to get stuck for a day in Comodoro, so bring a good book since NOTHING is happening there (unless you're a fan of the petrol industry, in which case you can visit their museum). Also, the connection from Rio Gallegos to Ushuaia will probably just leave as you arrive, which will cost you another day. In Rio G, this is not so bad, since it's a quaint, friendly town.

good luck, let us know how your trip has been,

Ive done from Ushuaia to El Calafate to Rio G. to Comodoro Riv and then to El Bolson. The standard of buses isnt as good as elsewhere in Argentina and it can be cold at night so dress warmly. Also, bring some snacks and drinks on board with you! Its a lllong route, but I enjoyed it. Countryside didnt change much on the east of Argentina but was still worth seeing in my opinion. Better than flying.

It's a long long way from El Bolson to El Calafate, though...
