Definitely go to Fraser Island, its beautiful- Huge stretches of golden sandy beaches and its so much fun rocking and rolling in a bus on the soft sand roads.

Snorkling is amazing at the Whitsundays (better than the barrier reef by far).

If you want a city - I'd recommend Melbourne as its laid back, theres loads to do and some great parks. It's basically got everything you could want.

The whitsundays are actualy islands in the Great Barrier Reef or surrounded by the reef, so diving in the Whitsundays is diving in the reef. There are over 74 islands to choose from.

For a city, sydney has more to offer than melbourne(in general)

I did a tour from Cairns to Sydney and it was great.

Fraser Island is amazing, really really beautiful, and I only spent 1,5 day there.
The Whitsundays are beautiful too, but living on a boat for 2 days wasn't really my thing, but there are 1 day sails available too.

And definately visit Sydney, awesome city!

Australia is awesome in general, absolutely loved it there!

I can't narrow it down to just one thing that you must see. But my absolute favorite things were:

Fraser Island
Cape Tribulation
Barossa Valley
Ayers Rock, and even better The Olgas
Snorkelling in Exmouth
Whitehaven Beach (in the Whitsundays)

You can't see them all unless its a big trip, so which ever of those are closest too, don't miss them.

West coast was amazing. And doing it with Wicked campers was really fun and worry free!
Monkey mia was great- def go back. It is only a small resort but we managed to get volunteer work with dolphins, which was an amazing experience. And people there are great!!!
Also I would say Darwin is the best city. Small but pretty and all shops, bars , food etc close by.
Haven't done much of east coast but looking forward to frasier island and whitsundays (esp after other commments! thanks):)

I forgot to mention Cape Tribulation! If you're up for some beautiful rainforest scenery, it's definately worth it. It's pretty deserted up there, but if you travel with a bunch of people it's very cool. We stayed at the Ferntree Rainforest Lodge, which is a nice resort.
You should also go swimming in Mossman Gorge while you're there!

In my opinion Tasmania is the most beautiful of all the states and should not be missed. I flew with Jetstar from Melbourne to Hobart for $100 return. (www.jetstar.com.au)

Difficult to pin down to one thing but if I had to it would be Kuranda, take a trip up to it from Cairns by train, visit the stalls, take in the bits of aborigine culture, do an army DUKW tour then take the skyrail(a cable car system) back down to Cairns - spectacular views of the rainforest(you can reverse the skyrail - train bit if you wish)
Of course you'd miss a visit to Uluru or the Great Barrier Reef, or the scenic railway in the Blue Mountains, not forgetting a pylon climb in Sydney, all things I enjoyed. :)
