Santiago, Chile, to Caracas in one go? That's one hell of a bus journey...

It can surely be done; the quickest by far would be to follow the pacific coastline as much as possible: Santiago - Iquique - Lima - Piura - Guayaquil - Quito - Tulcán - Bogotà - Cúcuta - Caracas. I mentioned border stations, since taking national busses is cheaper than international ones, but of course there's also direct service from Santiago to Lima to Quito, for instance.

Supposing a minimal waiting time when changing, this would still take you at least 4 days, though, and I think even that may be a bit too optimistic. Plus the pacific coast isn't exactly the most interesting part of SA...

Two weeks still is an awfully short time for so great a distance; yet, if you don't mind sitting on your butt for days on end staring out of bus windows, you can make this trip a little more interesting, scenery-wise.

What you could do, for instance, is go through Bolivia; Santiago - S. Pedro de Atacama - Uyuni - Oruro - La Paz - Copacabana - Puno - Machu Picchu/Cusco - Lima - Piura - Cuenca - Baños - Quito - see the other itinerary for the rest.

Mind you, this is crazy, but you might just pull it off in 14 days. If you don't stop in between, that is :)
