There are all kinds of possibilities - What kind of time frame are you thinking of? Are there any places you are really interested in seeing? Is there anywhere you might find less inspiring to go? I'm assuming from the post that you are driving, is this true? With a little more info we'll probably all be a little more helpful

So many places to visit, but the best way of travelling around would definately be by car and renting one is very easy and best done at home, where prices are better (for some strange reason). Avis, Hertz and Budget have good rates and are very good options with new cars.
I have only been to Ontario (canada) which is 'around the usa', but I have been reading and doing some research about good routes through the USA and Canada (going back to canada this september).
First of all, naturewise, I would really choose the western half of the continent, with the Rocky Mountains, many national parks, deserts and other natural wonders.
Some suggestions: from Denver to San Fransisco through the states of Colorado, Arizona, Utah, New Mexico, Nevada and California, with a possible detour to Montana/Idaho/Wyoming.
Or along the east coast from California up to Seattle. Or why not visit Alaska, possibly in combination with Yukon, Canada? For a more tropical adventure Florida is nice.
Not to forget Route 66 which partly is runs through the states of the first route I mentioned but starts in Chicago. The specific route itself is difficult to follow but that really doesn't matter.

Besides car travel, amtrak has some great train rides in the US as well, and again, from Chicago to for example LA, San Fransisco or Seattle should be great, maybe in combination with car rental as well.

I travelled across america using greyhound and amtrak. The Amtrak train from San Fransisco to Denver was gorgeous so much so i plan to do it again as part of a rtwt. Also from Denver (I never took to Denver) there isnt much distance to COlardo Rocky NAtional PArk for some great hiking.

I travelled earlier this year for a month using amtrak acros the usa, started in denver colorado and went to chicago, indianapolis, chicago, washtington, maryland and new york and then all the way back to denver, if you not to worried by time it was a enjoyable journey! Train was easy as the major stations were right in the middle of the city, not in the middle of nowhere. Some delays at times but generally all good, didn't bother me that much anyway as didn't have any set plans or schedules.
Saw some really great views on the train and met some nice people. Cheap ticket also which allowed unlimited travel for 2 months or a month..something like that.
