No, there is not any toilet in busses in Iran.
Not city busses and nor country busses.
In cities , its not any problem and even better that there is no toilet but for travelling around, sometimes it causes problems, but its so rare.
People know that there are no toilets so they care about their timing of loo and regarding to that they can wait with no problem until they reach a motel, a resturant or their destination.
I don't know if in tour busses there are toilets or not.

Iran uses a huge MERCEDDESS BENZ land fleet system for cities and Most Volvo and Mercedesses for out roads

The only thing better than Iranian buses are Turkish buses, which practically give you the feeling that you sitting in an airplane. Iranian Volvo buses are very modern and comfortable, that they have no toilets is generally no problem. The bus stops for prayers at rest stations, you can use the toilet there. (Be warned though that these toilets can be very icky - squat, no toilet paper and no water to flush either. So always go at your hotel.)
