Has anyone done Ushuaia- Puenta arenas on bus..One travel guides says there is a bus route form puenta Arenas to Ushuaia ,so I believe buses run on the reverse route as well.
I am sure it would be a journey laden with an amazing landscape of astounding beauty..If there is a website where I can book the tickets that would be awesome as my schedule is really tight and can't take chances by going theer and finding whether seats are available or not..and if someone has done it on bus..what time of the day buses generally depart and how long does it take to reach Puenta arenas..the guide says 12-14 hrs from PA to ushuaia

I am not sure how tight your schedule is, but if you arrive in Ushuaia you probably stay there for a night or two I guess?
In that case I wouldn't worry about buying tickets in Ushuaia as there are several bus companies that among them travel daily.
The trip is fantastic with great landscapes and although sometimes bumpy it never is boring. You will cross the Strait of Magelan by boat (bus goes on boat).
Most buses leave very early in the morning, like 5 or 6 AM.
Check this company for example, they leave 3 times a week, and it seems a bit later, 8 AM.
Or check this one Tecni Austral (the second in the list) travel there as well, you can email them.
