I want to hire a car in Melbourne and drive it to Sydney, is it cheaper to book from the U.K. or wait until I get to Melbourne, can anyone help? thanks stokesy

Doesn't really matter, as long as you don't go through an expensive middle man, I suspect (which'd undoubtedly be more expensive in the UK than in Australia).
That said, I ended up mostly booking my Australian rental cars through carhire.com.au, and it definitely made a huge difference there (with some car rental companies) if I didn't change the default "I live in" selectbox away from Australia. The rates offered to people from Australia were significantly cheaper (again, with some car rental companies; which (imo) totally sucks).
Of course, no one anywhere ever checked the value of that box after the booking was made, so I tended to just conveniently forget to change that field. :)

To add:
If you mean booking online with Hertz while you are at home in the UK, compared to arriving at the Melbourne Airport and going to Hertz to check prices: definately book in the UK!
It is mostly cheaper to book rental cars before arrving. In general that is. Of course, there are some local companies probably offering even better deals, but as a regular rule, booking online is cheaper than checking rates at the desk.
Same as with hotels most of the times...not always.
