I'm hoping to fly into Chicago for the start of April before visiting a friend in Birmingham, Alabama for Easter (a massive trek, I know!). I have looked into an Amtrak rail journey but this rather convoluted as it takes me to DC then Birmingham totalling 40+ hours. So I was hoping some knowledgeable folk might be able to point me towards some reputable bus companies or a more direct rail route?

Take a look at Greyhound. They are certainly the largest bus company in the US. Good luck.
Definitely take a look at Greyhound. Amtrak suffered track damages due to hurricane Katrina that have not been repaired yet. Their routes take you way out of your way - seriously. Once you could go from New Orleans directly to Jacksonville. Now, it's New Orleans to Washington DC and then back down to Jacksonville. The bus will get you where you want to go in less time and for less money. Amtrak is dealing with all sorts of problems due to natural disasters lately so they have had to alter their routes - most which are not convenient for long distant travelers.
