In Turkey you can move around where ever you want with buses. So until the very edge of turkey close to syria. But if you ask a direct bus service from Ist. to Damascus, I don't know! Some big bus companies serving almost all around turkey are Ulusoy, Varan and Kamil Koc, with their web sites and call centers, please check them.
Have a nice trip..

Having travelled by bus in Turkey and having gone by bus from Dublin to Germany and from Germany to Kiev, the only word I have to describe the idea of travelling from Istanbul to Turkey by bus is "completely mental".

First you need to realize that Turkey is a big country. Roads, especially in the east at the borders to Syria, Irak and Iran, are not the type you are used to in Western Europe or the U.S.. Mudslides are frequent in Winter and then even bigger highways are blocked for days. The buses are old too and going over the montains (3000 meters high) take a long time to complete what looks like a short journey on the map.

If you are really going, expect to spent 4-6 days in a bus with no toilet on roads where your bus is the only thing for miles on end and medical help won't be coming for hours or days if there is an accident. (Which are frequent, as the wracks beside the roads testify to.) The very steep ravines, the deserted huts of shepherds long emigrated to Germany and the rocks falling of cliffs down on the road don't inspire confidence either.

IMO, your best option really is to book a flight with Turkish or Syrian Airlines if you want to go from Istanbul to Damaskus.

However, if want to travel around Turkey and then cross the border, you might want to look into the option of going by train. There is a weekly train coming from Istanbul crossing the Turkish-Syrian border and going on to Damaskus. It should be possible to hop on that train along the way. Be warned that it is going to cost you.

Have a nice trip.
