Byron Bay is a must, for the beach and the town.
Nelson Bay is a pleasant place - a little quiet, but not bad for a stop over and dolphin/whale cruises etc.

I actually did the reverse on the weekend. If you are going to take your time maybe aim to stop in Coffs Harbour... its about half way and a pretty nice spot... Maybe on your first day stop at Port Stephens/Nelson Bay for lunch and power on from there... stay the night in Coffs and spend a night in Byron Bay... Surfers is only a coupleof hours from there so you could afford to sit on the beach for most of a day...

These suggestions are all assuming you are taking the Pacific Highway up there (which I prefer, it is a better road and much faster)... If you take the New England Highway there are also some nice small country towns to take in... personally I would go the coast road...
